چکیده
Actinic Keratoses (AKs) are premalignant intraepithelial skin lesions that can progress to cutaneous squamous cell carcinoma (SCC). The field of cancer immunotherapy has demonstrated the ability immune system treat late-stage cancers. We have shown also be leveraged prevent through a randomized, double-blind clinical trial. Topical calcipotriol plus 5-fluorouracil (5-FU) effectively eliminates AKs, leading SCC prevention in immunocompetent patients. Calcipotriol 5-FU activates robust CD4+ T helper 2 (Th2) immunity by inducing Thymic Stromal Lymphopoietin (TSLP) cytokine and stress signals keratinocytes. However, mechanism which activated Th2 cells eliminate AKs remains unknown. Herein, we investigated effector antitumor induced skin. RNA sequencing on trial samples revealed interleukin 24 (IL-24) was highly upregulated treated with compared alone. cell-derived cytokines, IL-4 IL-13, IL-24 expression lines. Intracellular induction lines resulted their apoptosis. To further investigate role downstream immunotherapy, developed Il24 knockout mice subjected them carcinogenesis protocol, mutant p53 clones resembling humans. Importantly, tumor protective effect markedly diminished mice. These findings reveal essential cell/IL-24 axis mediating function

Randomized trial of calcipotriol combined with 5-fluorouracil for skin cancer precursor immunotherapy.
BACKGROUND Actinic keratosis is a precursor to cutaneous squamous cell carcinoma. Long treatment durations and severe side effects have limited the efficacy of current actinic keratosis treatments. Thymic stromal lymphopoietin (TSLP) is an epithelium-derived cytokine that induces a robust antitumor immunity in barrier-defective skin. Here, we investigated the efficacy of calcipotriol, a topical...
متن کاملTopical 5-Fluorouracil associated skin reaction
Topical 5- Fluorouracil (5-FU) is used more frequently to treat actinic keratosis. We are presenting a skin reaction as a side effect of this medication. Treatment for such cases of 5-FU-induced skin reactions is based on proper skin care and treatment of any superimposed infections. Medical providers should be aware of such side effects to provide their patients with proper instructions to avo...

متن کاملTargeting the T Helper 2 Inflammatory Axis in Atopic Dermatitis.
Atopic dermatitis (AD) is a chronic inflammatory skin disease that affects up to 25% of children and 10% of adults. The skin of patients with moderate to severe AD is characterized by significant barrier disruption and T helper 2 (Th2)-driven inflammation, which are thought to play a significant role in the pathogenesis of AD. Current management of AD is aimed at suppressing the inflammatory re...
